چکیده
Abstract Background CD4+ T cells are associated with pulmonary arterial hypertension (PAH) pathogenesis but mechanistic insights limited. Purpose To identify differential methylation signatures in healthy controls vs PAH and evaluate a putative association the cardiopulmonary hemodynamic profile of affected patients. Methods We used RRBS platform to DNA methylome CLEOPAHTRA clinical trial. Results Differentially methylated CpG sites (N=631) annotated N=408 genes (DMGs). Most them (65%) were hypermethylated localized distal intergenic (36%) promoter regions (31%). Promoter-related network analysis established subnetwork highlighting 5 hub DMGs (SOCS3, GNAS, ITGAL, NCOR2, NFIC) non-hub (NR4A2, GRM2, PGK1, STMN1, LIMS2) as potential candidate (Figure 1). The Infinium Human MethylationEPIC BeadChip on from an independent study population confirmed global RRBS-methylation trends. Both idiopathic Associated-PAH, each these 10 network-oriented was strongly correlated at least one parameter such right atrial pressure (RAP), cardiac index (CI), mean (mPAP), vascular resistance (PVR), capillary wedge (PCWP) 2). In addition, mRNA levels NFIC, PGK1 IL-6-STAT3-SOCS3 signaling axis significantly upregulated PBMCs patients suggesting drug targets. Furthermore, both SOCS3 positively (CI) whereas RAP inversely CI Associated non-invasive biomarkers. Conclusions This hypothesis-generating shows for first time that circulating signatures, inclusive SOCS3, may yield insight into pro-inflammatory mechanisms exacerbate remodeling suggest biomarkers optimize patient phenotyping and, possibly, prognostication PAH. The Importance of Electrocardiography in Pediatric Patients with Pulmonary Arterial Hypertension in Follow- up
Background: Right Ventricular (RV) hypertrophy is an adaptive response to chronic RV pressure overload in patients with pulmonary hypertension. We investigated the relationships between RV hypertrophy indicators, including electrocardiography, the percentage oxygen saturation (SaO2%), body mass index (BMI), and blood uric acid levels in patients with...
متن کاملSex and Hemodynamics in Pulmonary Arterial Hypertension
Introduction: Female sex is a risk factor for pulmonary arterial hypertension (PAH), yet women have better survival than men. We sought to determine if sex was associated with baseline hemodynamics in subjects with PAH, and whether age modified these relationships. Material and Methods: We conducted a pooled analysis from 11 randomized trials submitted to the U.S. Food and Drug Administration. ...
متن کاملEvaluation of Circulating Proteins and Hemodynamics Towards Predicting Mortality in Children with Pulmonary Arterial Hypertension
BACKGROUND Although many predictors have been evaluated, a set of strong independent prognostic mortality indicators has not been established in children with pediatric pulmonary arterial hypertension (PAH). The aim of this study was to identify a combination of clinical and molecular predictors of survival in PAH. METHODS This single-center, retrospective cohort study was performed from chil...
متن کاملEvaluation of Pulmonary Hypertension In First Degree Relatives Of Patients With Primary Pulmonary Hypertension
Background: Pulmonary hypertension (PH) was defined for the first time in 1951 as primary pulmonary hypertension (PPH). Some studies emphasized on the role of genetics in the development of pulmonary hypertension in family members of affected patients. So, in this study we evaluated the prevalence of pulmonary hypertension in first degree family of patients with documented PPH. Methods: In thi...

Pulmonary arterial hypertension (PAH) is a progressive disease with high morbidity and mortality rates. Research has shown that PAH has a prevalence rate of 10-79% in thalassemia major patients. This cross-sectional study was carried out in 2014 to determine the prevalence and risk factors of PAH in all thalassemia major patients of over 18 years of age in Ilam, Iran. A cardiologist measured sy...
